Ancestry by DNA

The Ancestry by DNA test determines your bio-geographic origins. The test will give you your ancestral estimates of the four main population groups: Sub-Saharan African, European, Native American and East Asian. There has been migration between all the populations over a long period of time but these population groups are peoples that share a common ancestry.

This test is not attempting to define your racial background, the DNA Ancestry test will give you a statistical estimate of your BioGeographical Ancestry (BGA). BGA is a means of reporting the proportional ancestry of a person that is independent of the various socially determined “races.” While race is additionally defined by cultural, political and social conventions (there is no genetic definition of “race”), biogeographical ancestry describes groups of people that share similar DNA markers because of a shared heritage, which may span a number of geographical locations.

Scientific evidence tells us that the human species is fairly young. According to most archaeologists we originated in east Africa 130,000 to 400,000 years ago and then split into groups, multiplied, migrated, and settled in various locations across the world. Throughout this process and in the intervening time there has been significant independent evolution of the groups that established the various continents of the world. The evolution can be identified in the differences in allele frequencies at genetic markers. Predominantly, we see that markers found in one group are also prevalent in all groups and the markers that are the most common in one are also common in others.

You will be provided with a sample collection kit. Samples are collected using very simple and painless mouth swabs that are gently rubbed on the inside of the cheek. This method collects loose cells inside the mouth and it is these cells that we test.

In order to calculate your approximate ancestral origins we use specific genetic markers called Ancestry Informative Markers (AIMs) which are dissimilar between the groups and therefore indicate relative prevalence of each group in your make-up.

This test gives an estimated percentage of ancestry from four population groups:

  • Sub-Saharan African: Population group of people with roots in the Sub-Saharan region of Africa
  • Indigenous American: Population group who migrated to inhabit North, South and Central America
  • European: Includes Europeans, Middle Easterners and South Asians
  • East Asian: Includes Japanese, Chinese, Koreans and Pacific Islanders

You will receive the following:

  • Ancestry Test Certificate – a personalised certificate listing your ancestral ethnicity percentages
  • Results Manual – a detailed manual (PDF on CD) that explains your results as well as provides more information about ancestry testing and human migration history

Maternal Test

We are able to provide maternal lineage testing because the mitochondrial DNA is passed from a mother to all of her children and from her daughters on to their children, generation after generation. The mitochondrial DNA is very stable and is inherited with relatively little change over multiple generations. Because mitochondrial DNA is common to everyone, this test can be done for both men and women.

Hypervariable regions in mitochondrial DNA are the sites where mutations occur and it is these that we test and this way we can designate your haplogroup. On average one letter in the sequence of mitochondrial DNA changes every 1,500 years. This change is then passed on to succeeding generations.

Using this important part of your DNA, your maternal history can be traced back to the first individuals who populated the planet. Scientific data suggests this was in Africa over 130,000 to 400,000 years ago.

The modern human was born somewhere in East Africa approximately 175,000 years ago. From this base, people began to migrate out of the African continent to settle in other parts of the world. Over time as they started to settle in different geographic areas, their DNA took on changes until they had distinct differences and it is these distinct genetic differences that allows us to define to which haplogroup you belong and which place of origin you are associated with.

There are three parts of the mitochondrial DNA that can be examined during the testing process:
HVR1 – testing this area allows us to determine your haplogroup
HVR11 and HVR111 – testing these areas assist with the designation of the haplogroup
and in some cases the sub-group known as a sub-clade.

You will receive the following:

  • Your Family’s Ancestral Map – a map showing the journey your ancestors took including where they first settled in the ancient world.
  • Your Haplogroup Designation – confirmation of your haplogroup plus information about it including where peoples with your haplogroup are most commonly found
  • Your MTDNA Raw Data – this lists the differences between your sequence and that of the revised Cambridge Reference Sequence
